During this period, the main antagonist, Muzan Kibutsuji, was born. He was initially a human receiving treatment for an incurable disease. In frustration over the lack of progress with his treatment, Muzan killed the doctor and consumed him. This act transformed him into a demon, granting him increased power. He discovered that sunlight would kill him, and thus began researching to obtain immortality.

During the Sengoku era, the Demon Slayer Corps experienced a golden age. This period saw the rise of powerful swordsmen such as Yorichii and Michikatsu. Most of the scenes in the Demon Slayer anime take place during this period. Everything from Demon Slayer season 1, Mugen train to Swordsmith Village Arc happened in this period. Muzan Kibutsuji was killed in this period, and the Hashiras became more powerful with their unique breathing styles. The twelve Kizuki demons were also killed during this time, including Upper Moon Three, Upper Moon Two, and Upper Moon One during the Infinity Castle arc.
